B21: A Dedicated Solution State SAXS Beamline
Diamond offers a dedicated solution state Small Angle X-ray Scattering (SAXS) beamline, B21, capable of accommodating a wide variety of aqueous-based samples and some organic solvent-containing samples. To achieve high sample throughput, B21 is equipped with a highly automated BioSAXS robot for small volume liquid handling from a 96-well plate.
BioSAXS Robot
The BioSAXS robot enables scientists to perform high-throughput solution scattering, processing each sample in just 5 minutes. Utilising 96-well plates, which we can supply, or PCR strips if preferred, the robot delivers samples to a fixed glass capillary inside the exposure unit, held under vacuum to minimise background noise. Samples can be refrigerated while awaiting data collection, ensuring their integrity.
Integration of SEC-SAXS
Diamond has developed a high-performance liquid chromatography (HPLC) sample environment for solution scattering (SEC-SAXS). This integration couples established biological separation techniques with the scattering experiment, effectively removing impurities and protein aggregation to enhance the quality of the scattering curves.
BioSAXS Lab
Our dedicated BioSAXS lab is equipped to meet all your sample preparation needs. We offer storage for your samples under appropriate conditions, including refrigeration, -18°C freezer, and -80°C freezer. Additionally, we can perform HPLC experiments to separate sample components, ensuring high-quality data.
Software
The Scatter software, partially developed at Diamond, is available for download and offers a range of analysis options. It is freely available for industrial users, with the latest version included in each B21 experiment folder. Please note that a license for ATSAS is required to use some additional add-ons.
